Comparison of haemocytic parameters among flat oyster Ostrea edulis stocks with different susceptibility to bonamiosis and the Pacific oyster Crassostrea gigas ArchiMer

Farming of the flat oyster Ostrea edulis in Europe is severely constrained by the protozoan Bonamia ostreae. The introduction of the resistant species Crassostrea gigas has been a relief for the farmers, while the pilot programmes to select O. edulis strains resistant to bonamiosis performed in various countries can be seen as a promising strategy to minimise the effects of bonamiosis. However, the physiological bases of this differential susceptibility remain unknown. A search for an explanation of the intra and interspecific differences in oyster susceptibility to bonamiosis was accomplished by comparing some immune parameters among various O. edulis stocks and C gigas. The mytiliculture is one of the most important aquaculture in the world, however, little is known on the mussel immune system, which plays a critical role in case of infectious diseases. Like all the other invertebrates, our model, the Mediterranean mussel Mytilus galloprovincialis, possesses only an innate immune system considered to be primitive and non-specific. Effect of diets containing different levels of highly unsaturated fatty acids on physiological and immune responses in Pacific whiteleg shrimp Litopenaeus vannamei (Boone) exposed to handling stress ArchiMer

Juveniles fed a diet containing a low or a high level of highly unsaturated fatty acids (HUFA) for 38 days were exposed to handling stress. In a first experiment, stress was applied daily for 30 days, after which the physiological and immunological variables were measured, whereas in a second experiment, stress was applied once and samples were obtained 1 and 24 h after the stressor event. Shrimp that were stressed for 30 days showed significantly lower survival, final weight and feed consumption compared with unstressed shrimp. The concentration of the high-density lipoprotein beta-glucan-binding protein was significantly higher in shrimp fed the high-HUFA diet. Effects of temperature and salinity on haemocyte activities of the Pacific oyster, Crassostrea gigas (Thunberg) ArchiMer

The Pacific oyster, Crassostrea gigas, is extensively cultivated and represents an important economic activity. Oysters are reared in estuarine areas, subjected to various biotic and abiotic factors. One of the limiting factors in aquaculture is mortality outbreaks, which may limit oyster production, and the causes of these outbreaks are not completely understood. In this context, the effects of temperature and salinity on Pacific oyster, C. gigas, haemocytes, were studied. Haemocytes are the invertebrate blood cells and thus have been shown to be involved in defence mechanisms. Flow cytometry was used for monitoring several haemocyte parameters. Penaeidins are members of a new family of antimicrobial peptides isolated from a crustacean, which present both Gram-positive antibacterial and antifungal activities. We have studied the localization of synthesis and storage of penaeidins in the shrimp Penaeus vannamei. The distribution of penaeidin transcripts and peptides in various tissues reveals that penaeidins are constitutively synthesized and stored in the shrimp haemocytes. It was shown by immunocytochemistry, at both optical and ultrastructural levels, that the peptides are localized in granulocyte cytoplasmic granules, The expression and localization of penaeidins were further analysed in shrimp subjected to microbial challenge.
